چکیده

Abstract Background Osteoarthritis (OA) is a chronic degenerative disease that suppresses middle-aged and older people worldwide. Silent information regulator 1(SIRT-1) associated with several age-related diseases, such as cardiovascular neurodegenerative diseases tumors, etc. The protective role of SIRT-1 in bone joint has become increasingly well known. Objective To explore the relationship between its related factors OA. Methods Fresh tibial plateau specimens were collected from 30 patients knee OA who underwent total arthroplasty. According to results Safranin O Fast Green Staining, hematoxylin–eosin staining OARSI grade developed by International Association for Study Osteoarthropathy, divided into mild group, moderate group severe damage cartilage was evaluated. protein levels samples analyzed immunohistochemistry. Then, take 60 8-week-old female C57BL/6 J mice apply Destabilization medial meniscus (DMM) induce Mice randomly normal (sham), model (model), post-modeling drug administration (srt), each further 2 weeks after modeling (2 W) 8 (8 according time surgery. degree mouse evaluated using Staining grade. Immunohistochemical techniques assessed SIRT-1, β-catenin, LEF-1, MMP-13 Collagen II samples. LEF-1 immunohistofluorescence technique. mRNA expression real-time quantitative polymerase chain reaction (qPCR). Results In human samples, compared showed layer structure, number chondrocytes decreased, cell hypertrophic, surface discontinuous, increased. had injury highest immunohistochemical analysis, W groups significantly increased than sham groups. decreased ( P < 0.05), srt group. immunofluorescence 2w 8w qPCR results: Compared groups, while contrast, Conclusion SRT-1720, specific activator does increase level SIRT-1. may play regulating inflammatory
